Pfu enzymes can catalyze the polymerization of deoxynucleotides in the 5' to 3' direction, depending on the DNA template. Unlike Taq enzymes, Pfu enzymes have a 3' to 5' proofreading activity. In addition, the Pfu enzyme has 5' to 3' exonuclease activity, but the Pfu enzyme has no reverse transcriptase activity.
